Product Description

Norgen’s Plasmid MiniPrep Kit enables rapid analytical scale preparation of high quality plasmid DNA.The kit is able to purify plasmids up to 13,000 bp in size. Each column has a binding capacity of 25 µg, and the typical purification yield is up to 20 µg from 1.5 mL of bacterial culture.The purified DNA is free of genomic DNA, RNA and cellular proteins and is suitable for many downstream applications.

Purification is based on spin column chromatography. Briefly, thebacteriaharbouring the plasmidis pelleted and lysed, and the released plasmid DNA isbound to Norgen's column in the presence of Binding Solution (BIND).Under these conditions only the DNA will bindtothe columnwhilemost ofthe contaminating RNAand cellular proteinaceous componentsare removed in the flowthrough. The bound DNAis then washed to remove anyremaining impurities(WASH).Lastly, the purified DNA is eluted into 50 - 100µL of the provided ElutionBuffer or water(ELUTE).Pleaseseeprocedure flowchart to the right.

This kit is also available in a 96-well format for high-throughput plasmid purification.Purification with the 96-well plates can be performed using either a vacuum manifold or centrifugation.

Figure 1.High DNA Yield from Small Cultures. Plasmid DNA from 1 mL overnight bacterial cultures was prepared with Norgen’s Plasmid MiniPrep Kit and compared with two other commercial kits.The purifications were performed in triplicate, and the average yield for each kit is shown in the graph. The quantification of the DNA yield was performed by resolving 5 µL of the 50 µL of eluted DNA on a 1X TAE, 0.9% agarose gel followed by densitometry. Norgen shows a significantly higher average yield than the other 2 competitor’s kits.

Figure 2.High-Accuracy Sequencing. One microgram of plasmid DNA purified by Norgen’s Plasmid Miniprep DNA Kit was used as a template in an Applied Biosystem DNA Sequencer. The result showed an accuracy of >99% over a 950 bp contiguous sequence.

Storage Conditions and Product Stability
All solutions should be kept tightly sealed and stored at room temperature. These reagents should remain stable for at least 1 year in their unopened containers. The RNAse should be stored at -20°C upon arrival. The Resuspension Buffer should be stored at 4°C upon addition of RNAse enzyme.
